摘要:
为了选育稳定高效的茧丝脱胶菌株,文章首先利用PCR-DGGE技术对茧丝腐化液微生物群落结构进行了解析,研究结果表明,腐化液的优势菌属为芽孢杆菌属(Bacillus)。对腐化液中脱胶微生物进行了筛选,获得了一株脱胶率较高的菌株,经16S rDNA分子鉴定,命名为Aeromonas sp. TJ-5。对脱胶菌株脱胶条件进行了优化,获得了最适的pH值、温度和接种量分别为pH 7.0、37℃和3%。在优化的培养条件下发酵48 h,脱胶率达到28.2%,蛋白酶活力为59.5 U/mL。研究结果为建立茧丝的微生物纯种发酵工艺奠定了基础。
Abstract:
In this study, to screen stable and high-activity strains for silk degumming, microbial community structure of silk natural fermentation liquid was firstly investigated by PCR-DGGE technology , and the results showed that the dominant genus was Bacillus. Screening of degumming microorganism possessing from silk natural fermentation liquid was performed, and a bacterium with high degumming rate was obtained. The bacterium was named Aeromonas sp. TJ-5 based on 16S rDNA molecular identification. The degumming conditions of TJ-5 were optimized, and the optimal pH, temperature and inoculum concentration were pH 7.0, 37 ℃, and 3%, respectively. After cultivation for 48 h under the optimal conditions, the degumming rate and protease activity reached 28.2% and 59.5 U/mL, respectively. These results may contribute to developing a new degumming process for pure microbial fermentation.
